Rock 4 Regal will return after the success of last year's event, which raised more than $3000 for MND research.
The charity concert is in honour of Adam Regal and his Regal Regiment.
The former Kiama Downs resident learnt that he had the disease in December 2013.
For the most part, the illness has progressed slowly for the father of two.
However, the Flinders resident has already lost two members of his immediate family to the disease.
A Wollongong production company recently created a national television commercial featuring Mr Regal for national Motor Neuron Disease (MND) Week.
Every cent raised from the concert through ticket sales, donations and raffles will be donated to the MND research team at Macquarie University to help in the search for a cure.
This year's event will include performances by Furious Monk, Arcane Saints (Victoria), The Vanns, Crash Tragic, Mixtape for the Drive, Roses In Hand and Tommy M and The Mastersounds.
Organiser Den Murray is a long-time friend of Mr Regal.
Murray, originally from Kiama Downs but who now resides in Melbourne said they hoped to better last year's total, even if it was only by one dollar.
"Local businesses have donated prizes for the raffle, so I'm pretty sure that will help us break that amount," he said.
"It's a really good, diverse line-up of bands, and they've all helped to promote it.
"It's awesome to see; they're not asking for money, and giving up their time for a good cause.
"We'll also have Rock 4 Regal shirts for sale."
Murray also plays drums for Arcane Saints, who recently released their In the Shade of the Juniper EP, and toured the US in 2014.
He joined the rockers about two years ago, and was enthused about returning home with his latest band.
"I don't think any of my mates (in the Illawarra) have seen this band yet, so that will be cool."
The over-18s event will take place on Saturday, June 6, at Kiama Leagues Club.
Doors open at 4pm.
Tickets are $10 plus booking fee, available from the oztix.com.au website and Haworth Guitars, or $15 at the door.
