The event will start at 8.30am with participants following a 4.2km figure eight loop to the lighthouse and back, then to Kendalls Beach and back.
Event organiser Julia Szulerowski said participants could walk or run anything from a single lap to a half marathon - it will not be a race and no times will be kept.
Strollers and wheelchairs are welcome. Money raised from the event will go to the Neurofibromatosis Association of Australia - neurofibromatosis (NF) is a genetic condition causing benign tumours to grow on nerves, bones, skin, the brain and the spinal cord.
Pressed against the optic nerve, they can damage sight.
There is currently no cure, but Ms Szulerowski hopes to raise awareness of the condition - last year's event raised $6000.
"It's one of the most common genetic disorders, but very few people know about it," she said.
Ms Szulerowski and her daughter, Jessica, were both born with the NF1 and have undergone countless operations between them to remove tumours.
Registration is $20 for adults, $15 for school-aged children, $45 for families and if participants wish to register a team, the cost is $20 per person.
A barbecue will be provided.
