The MS 24-hour Mega Swim will be held at the University of Wollongong Recreation and Aquatic Centre on March 22 and 23. Registrations are open.
The Figtree Lions hope to raise at least $45,000 this year with the community, Olympians McKeon and Poort and Commonwealth Games hopeful Emma McKeon.
The three have joined in previously and so far, the event has raised more than $120,000 for people living with MS. Previous stars included long-distance swimmer Robert Hurley, Wollongong triathlete Ben Allen and British triathlete and Xterra competitor Jacqui Slack.
Last year, more than 225 swimmers from 17 teams covered 1000 kilometres in the tag team relay-style Mega Swim and organisers hope even more people will take part in March.
The style of swim - freestyle, backstroke or doggy paddle - is up to the individual and swimmers of all abilities are welcome. Points are awarded to teams for every 500 metres completed.
Some entrants dress up in crazy costumes for the challenge.
